Reference gene selection for quantitative PCR studies in bovine neutrophils
نویسندگان
چکیده
Reference genes are essential for studying mRNA expression with quantitative PCR (qPCR). We investigated 11 candidate whole-blood neutrophil reference genes (ACTB, B2M, G6PD, GAPDH, GYPC, HPRT, PGK1, RPL19, SDHA, TFRC, and YWHAZ) for beef calves, both males and females, with or without selenium supplementation. Initial screening was based on gene expression level (<28 Cq cycles), variability (SD < 1.5 Cq cycles), excluded GYPC and TFRC from further analysis. Expression stability of the remaining genes was evaluated using four software programs: geNorm, NormFinder, BestKeeper, and the comparative delta Cq method. The neutrophil reference genes, YWHAZ, PGK1, and RPL19, consistently ranked among the top four most stable genes under these experimental conditions. The commonly used reference genes, ACTB and HPRT, were not reliable, underscoring the need to validate neutrophil reference genes under different experimental conditions. Multiple reference genes rather than a single gene may provide more robust and reliable results. The best pair of reference genes in whole-blood neutrophils from beef calves overall was PGK1|YWHAZ.

Selection of suitable reference genes for real-time PCR studies of early developmental stages of sturgeons
In quantitative real-time PCR, the mRNA level can be quantified in relative terms based on the expression ratio of mRNAs of the target gene and an internal reference gene. Since, an internal standard should be expressed at a constant level among different tissues of an organism at all stages of development, and should be unaffected by the experimental treatment, the stability of different refer...

In the present research, molecular detection of bovine leukocyte adhesion deficiency (BLAD) and complex vertebral malformation (CVM)in a population of Iranian Holstein cows has been carried outusing milk somatic cells by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP). The BLAD and CVM are monogenic and autosomal recessive heredity lethal syndrome in Holstein-Friesi...

The use of polymorphic markers in breeding programmes could make selection more accurate and efficient. A total of 324 individuals from six Iranian cattle populations (Sarabi, Golpayegani, Sistani, Taleshi, Mazandarani, Dashtiyari), F1 Golpayegani × Brown Swiss and Iranian buffalo populations were genotypedfor the Pit-1 HinfI and leptin Sau3AI polymorphisms by the polymerase chain reactio...

متن کاملSelection of Reference Genes for Quantitative Real-Time PCR during Flower Development in Tree Peony (Paeonia suffruticosa Andr.)
Tree peony (Paeonia suffruticosa) is a perennial plant indigenous to China known for its elegant and vibrantly colorful flowers. A few genes involved in petal pigmentation have been cloned in tree peony. However, to date, there have been few studies on the comparison and selection of stable reference genes for gene expression analysis by quantitative reverse-transcription PCR (qRT-PCR) in this ...
